Zusammenfassung:	The invention discloses an investigation method for fish biodiversity in a natural ecological environment. Echo detection and environment DNA technologies are adopted for investigation and sampling; comprehensively setting a survey route of a ship according to the environment, setting a water body sampling point, and processing, analyzing and detecting data; and comprehensively calculating the biodiversity index through detection data and an environmental DNA sample detection result. Compared with the prior art, qualitative and quantitative biodiversity data acquisition of fishes can be synchronously completed in one voyage, the investigation working time and cost are remarkably saved, and the method has the remarkable advantages of being rapid, efficient, free of damage to biological resources and the like; according to the method, the individual quantity and biomass data of each type of fish in the monitored area can be obtained, and the data are core data necessary for further calculating various biodivers
