The invention discloses a preparation method of a transgenic sterile chicken, and belongs to the field of gene engineering. The method comprises the following steps: (1) inserting an HSV-TK gene into the 3'end of the last coding exon of a chicken primordial germ cell specific expression DAZL gene at a fixed point by using CRISPR/cas9 mediated homologous recombination repair; enabling the primordial germ cells to specifically express the HSV-TK gene, and reinjecting the HSV-TK gene into a receptor chick embryo to prepare an HSV-TK chimera chicken; and (2) hybridizing the HSV-TK chimera chicken to obtain an HSV-TK receptor chick embryo, and giving ganciclovir to eliminate endogenous germ cells to obtain a transgenic sterile chick embryo.
