The preparation method of the nano-porous graphene oxide chip comprises the following steps: placing graphene oxide in a chloroform solution of octadecyl ammonium chloride to form a graphene oxide/dioctadecyl dimethyl bromination solution which is marked as a GO/DODA solution, spin-coating the GO/DODA solution on the surface of a substrate to form a nano-porous graphene oxide substrate, and drying the nano-porous graphene oxide substrate to obtain the nano-porous graphene oxide chip. The preparation method comprises the following steps: sequentially carrying out silanization treatment and amidation treatment on a nano-porous graphene oxide substrate, then bonding streptavidin, and finally modifying an antibody to obtain the nano-porous graphene oxide chip.
