A method for rapidly synthesizing a self-enhanced electrochemical luminescence probe based on polylysine in a water phase comprises the following steps: dissolving Ru (bpy) 2Cl2, 2, 2 '-bipyridine-4, 4'-dicarboxylic acid and NaHCO3 in a methanol aqueous solution, and reacting to obtain Ru (II) crystals; the preparation method comprises the following steps: performing constant-temperature incubation on Ru (II) crystals, EDC and NHS in a carboxyl activation buffer solution environment to obtain a Ru (II)-NHS ester solution, performing mixed incubation on the Ru (II)-NHS ester solution and polylysine, and performing purification to obtain a Poly (Lys-co-Ru (II)) product; and carrying out antibody labeling on the Poly (Lys-co-Ru (II)) product to obtain the corresponding probe.
