Zusammenfassung: | The invention provides a method and a kit for interaction analysis of protein and DNA (Deoxyribose Nucleic Acid) under an acidic condition. The method comprises the following steps: in the presence of EDTA (Ethylene Diamine Tetraacetic Acid), carrying out digestion reaction on deoxyribonuclease II (DNase II) and a protein-DNA interaction product under an acidic condition; stopping the digestion reaction, and carrying out enzymolysis reaction on protein in the digestion reaction liquid by using protease so as to dissociate DNA (Deoxyribose Nucleic Acid); according to a detection result of the free DNA in the enzymolysis reaction solution, determining the interaction size of the protein and the DNA. The DNase II is used for digesting different protein-DNA interaction products under the acidic condition, analysis of interaction of protein and DNA under the acidic condition and detection of the protection capability of protein on DNA under the acidic condition are achieved, and non-sequence specificity is achieve |
