Zusammenfassung: | The invention discloses a low-quality FFPE DNA treatment method, which comprises: (1) mixing DNA extracted from an FFPE sample with a thermal repair reagent, and carrying out thermal repair, the thermal repair reagent comprising a lysis buffer for extracting nucleic acid and protease K; (2) purifying the material obtained in the step (1) to remove the unreacted thermal repair reagent in the material so as to obtain thermal repair DNA (Deoxyribose Nucleic Acid); (3) the hot repair DNA is mixed with an enzyme repair reagent for enzyme repair, and the enzyme repair reagent has the functions of deamination repair, notch repair, oxidized base repair and 3 '-terminal closed repair; and (4) purifying the material obtained in the step (3) to remove the unreacted enzyme repair reagent, thereby obtaining the product.
