Firstly, it is found that replication of small RNA viridae virus FMDV can be promoted by inhibiting KLHL34 gene expression in host cells; secondly, the invention provides sgRNA specifically targeting KLHL34, the sgRNA can specifically target the KLHL34 gene, the knockout of the KLHL34 gene is realized by combining a CRISPR-Cas9 technology, and the obtained monoclonal cell line can significantly promote the replication of the small RNA viridae virus FMDV, improve the production quantity and antigen yield of the FMDV vaccine, and can be used for the preparation of the FMDV vaccine.
