The flanking sequence provided by the invention comprises a right flanking sequence as shown in SEQ ID NO. 1 and a left flanking sequence as shown in SEQ ID NO. 2. The invention provides specific primers SEQ ID NO. 9 to 10 or SEQ ID NO. 11 to 12 for detecting the flanking sequences. The specific primer provided by the invention is used for carrying out PCR (Polymerase Chain Reaction) amplification on a DNA (Deoxyribose Nucleic Acid) sample of rice to be detected, so that whether a T-DNA fragment of pC0309-KhvMaauMCMK5400 is inserted into the 32791293 to 32791426 basic groups of a non-coding region of the No.4 chromosome of the rice or not can be specifically indicated.
