The flanking sequence provided by the invention comprises a right flanking sequence as shown in SEQ ID NO. 1 and a left flanking sequence as shown in SEQ ID NO. 2. The invention provides specific primers SEQ ID NO. 9 to 10 and SEQ ID NO. 11 to 12 for detecting the flanking sequences. The specific primer is used for carrying out PCR (Polymerase Chain Reaction) amplification on a DNA (Deoxyribose Nucleic Acid) sample of rice to be detected, so that whether a T-DNA fragment of pC0309-KhvMaauMCMK5400 is inserted into a basic group from 19881585 to 19881622 of a sixth exon of the zinc finger protein Os09g0511500 of the No.9 chromosome of the rice or not can be specifically indicated. According to the invention, detection and safety management of the transgenic rice and the derivative line thereof are successfully realized.
