Zusammenfassung:The invention discloses an acquisition method and application of chicken primordial germ cells for strain resource breed conservation. An EAV-HP gene is inserted into the SLCO1B3 gene locus of the cell, the insertion sequence is shown as SEQ ID NO: 1, and the cell has a transient expression green fluorescent protein marker. The cell is obtained by the following method: separating gonads from embryos of a green-shell laying hen strain needing breed conservation, digesting by pancreatin, separating and purifying primordial germ cells by a differential adhesion method, and transferring into a green fluorescent protein gene through an adenovirus vector. The cell can be preserved in liquid nitrogen for a long time at low cost. The cells are microinjected into a Bailaiaviation chick embryo, a chimeric chicken can be obtained after hatching, and a green-shell laying hen strain living body needing breed conservation can be obtained again from offspring of the chimeric chicken.
