The method comprises the following steps: adding a premixed solution into a to-be-detected solution containing SAM for reaction, then adding a glycine oxidase detection solution for reaction after the reaction is finished, and detecting the generation amount of hydrogen peroxide in a reaction product to obtain the SAM content, through combination of glycine/sarcosine N-methyltransferase and glycine oxidase, the SAM content in the range of 0-2mM can be accurately detected on a 96 microwell plate, and the method is accurate in detection, proper in color development range and small in influence of other factors.
