Zusammenfassung: | The invention relates to the technical field of biochemical engineering, in particular to a method for expressing D-amino acid oxidase and application of the D-amino acid oxidase. Wild type D-amino acid oxidase with catalytic activity on a catalytic substrate DL-glufosinate-ammonium is obtained through screening, and the activity and soluble expression of the D-amino acid oxidase obtained through host expression are further improved through methods of screening expression hosts, adding molecular chaperones and the like. The method for preparing alpha-keto acid by using D-amino acid oxidase provided by the invention realizes kinetic resolution of racemic glufosinate-ammonium, and can be widely applied to production and application of L-glufosinate-ammonium.
