Zusammenfassung: | The invention discloses a method for quantitatively regulating and controlling gene translation and application thereof. The method comprises the following steps: sequencing the translation efficiency of a Kozak sequence before an initiation codon of a target gene or before an upstream open reading frame of a non-coding region of the target gene and a variant of the Kozak sequence, performing gene editing on the Kozak sequence as required according to a sequencing result, and realizing quantitative regulation and control of gene translation based on in-situ manipulation of the Kozak sequence. The lengths of the Kozak sequence and the variant of the Kozak sequence are respectively and independently 3-6bp. The invention establishes a gene expression regulation method which is efficient, flexible and extremely wide in applicability. A Kozak sequence of a target gene is customized in situ by using a precise gene editing tool, so that the expression level of the gene is quantitatively controlled in a gene translat |
