Zusammenfassung: | The invention discloses a multi-step purification process of human chorionic gonadotropin, which not only optimizes the purification process steps, but also redesigns a chromatographic column required by purification. Specifically, the inventor utilizes a cross-linked microsphere-gel chromatographic column and an affinity chromatographic column to sequentially separate and purify a human chorionic gonadotropin stock solution, and meanwhile, a filler of the cross-linked microsphere-gel chromatographic column is designed to be composed of 60-70% of sulfonic acid cross-linked microspheres and 30-40% of chlorinated silica gel. A ligand of an affinity chromatography column is designed to be composed of a mutated HCG receptor. The titer of the purified human chorionic gonadotropin pure product is about 9000 U/mg, and even can reach 10000 U/mg.
