Zusammenfassung: | The invention discloses a sphingomonas paucimobilis CRISPRi system and a construction method and application thereof, and belongs to the technical field of genetic engineering, the construction method comprises the following steps: 1) screening a vector capable of being stably replicated and expressed in sphingomonas paucimobilis; (2) replacing the replicon fragment of the plasmid plv-dCas9-sgRNA with the replicon fragment of the vector to obtain a recombinant plasmid pBBR1-dCas9-sgRNA, namely a CRISPRi (clustered regularly interspaced short palindromic repeats i) system; and 3) determining a target gene, designing sgRNA, and verifying the inhibition efficiency of the CRISPRi system. The CRISPRi system comprises an expression vector carrying an sgRNA expression cassette and a dCas9 gene, and the vector carries chloramphenicol resistance.
