Sheep induced pluripotent stem cell and preparation method thereof

The invention discloses a preparation method of sheep induced pluripotent stem cells, which comprises the following steps: constructing piggyBac transposon plasmids carrying four serially connected sheep-derived transcription factors, namely OCT4, SOX2, KLF4 and c-MYC and two serially connected sheep-derived transcription factors, namely NANOG and LIN28, and transfecting the piggyBac transposon plasmids into fetal fibroblasts of sheep through electroporation; and selecting clones of which the forms are similar to those of the embryonic stem cells, then screening cell clones conforming to the characteristics of the embryonic stem cells, and carrying out subculture and identification to obtain the sheep induced pluripotent stem cells.
