The invention provides a detection method of dimethylglycine in serum and application thereof, the detection method comprises the following steps: (1) mixing a to-be-detected sample with an internal standard solution, centrifuging to obtain a supernatant, and eluting through a column to obtain an eluted sample; and (2) mixing the eluted sample with a derivatization reagent and a reaction activator, carrying out a derivatization reaction, carrying out HPLC detection on the product, and calculating the content of dimethylglycine in the sample to be detected according to the detection result.
