The method comprises the following steps: acquiring a dark field microdroplet image, and extracting a foreground mark, a background mark and a gradient magnitude of the image from the dark field microdroplet image; superposing the foreground mark, the background mark and the gradient magnitude of the image to obtain a gradient magnitude image; and performing segmentation according to the gradient magnitude image to obtain an image of each droplet in the dark field droplet image.
