The in-situ online determination method comprises the following steps: step 1, obtaining an original spectrum library; step 2, extracting corresponding excitation wavelength and emission wavelength according to the three-dimensional fluorescence spectrum in the original spectrum library in the step 1 and the pigment type of the red tide algae, and constructing a standard spectrum library of corresponding discrete excitation-emission fluorescence spectrum; step 3, constructing a feature space for projecting feature information according to the standard spectrum obtained in the step 2; and step 4, inputting the discrete excitation-emission fluorescence spectrum of a sample to be detected into the feature space constructed in the step 3 to obtain corresponding feature information, performing predictive analysis on the feature information through the pre-constructed neural network, and outputting an identification result
