The nucleotide sequence of the promoter element is shown as SEQ ID NO.2 or SEQ ID NO.3. The promoter element has the capability of remarkably improving the detection sensitivity of metal Ni < 2 + >. The promoter element is used for transcriptional expression of NAD < + > synthetase and formate dehydrogenase genes to prepare an electrochemical signal output sensor, and by increasing the total amount of intracellular electron pools (NAD < + >) and the ratio of NADH < 2 + >/NAD < + >, output of electrochemical signals after induction of submarine metal Ni < 2 + > ions is achieved. The invention provides a new thought and method for screening an efficient inducible promoter and a detection mode, and improves the accuracy, safety and efficiency of metal Ni < 2 + > detection.
