Zusammenfassung:The invention discloses a kit for high-throughput determination of double-stranded DNA concentration and application thereof. The kit comprises a dsDNA dye, a microwell plate and a plate sealing film, the dsDNA dye is sub-packaged in micropores in the microwell plate; and the plate sealing film completely seals micropores in the microporous plate. The kit provided by the invention is applied to determination of dsDNA concentration. A method for measuring the concentration of the high-throughput dsDNA sample by adopting the kit comprises the following steps: 1) tearing off the sealing plate film of the kit; (2) respectively adding a dsDNA sample to be detected and dsDNA with known concentration into the dsDNA dye of the kit; 3) putting the microplate added with the dsDNA in the step 2) into a microplate reader for fluorescence detection to obtain fluorescence signals of the sample to be detected and the dsDNA with known concentration; and calculating the concentration of the dsDNA in the sample to be detected
