According to the invention, the complex grain rice gene CS is cloned for the first time, the gene encodes a BR degrading enzyme CYP734A4, a CS promoter in the complex grain rice contains insertion of 34 AT copies relative to a reference genome, the promoter activity can be enhanced, the CS gene in the complex grain rice specifically has up-regulated expression in pedicels and secondary branch meristem, and the specific expression of the CS gene in the complex grain rice is up-regulated in the pedicels and secondary branch meristem. Compared with non-compound-grain rice, the compound-grain rice has the advantages that the yield can be obviously increased by about 10% under the same background without changing the grain size and the plant height of the rice, and a powerful means is provided for high-yield breeding of the rice.
