The invention also discloses a separation and purification method of the lactobacillus sake ZFM225 bacteriocin, which comprises the following steps of: precipitating fermentation supernate of the lactobacillus sake ZFM225 by using an ammonium sulfate precipitation method to obtain crude protein; desalting the crude protein to obtain a protein crude extract; carrying out cation exchange chromatography separation on the protein crude extract, eluting with a gradient washing solution containing 0.5-1.0 M NaCl, and carrying out desalination treatment on the obtained eluent to obtain a protein primary purification solution; and carrying out reversed-phase high-performance liquid chromatography purification to obtain the lactobacillus sake ZFM225 bacteriocin.
