The preparation method comprises the following three main steps: primary culture, myoblast purification and subculture. An immortalized skeletal muscle myoblast line is established on the basis of muscle tissues on the back of the grass carp by adopting a collagenase and trypsin combination method, and confirmation is carried out through myogenic characteristic indexes and chromosome characteristic indexes of the grass carp. The cell line has good transfection performance, has remarkable growth performance difference under treatment of different arginines, and can be used for research work of regulation and control effects of specific gene functions and nutrient substances on grass carp muscle growth and differentiation, molecular mechanisms of the regulation and control effects and the like.
