According to the method, a titanium dioxide nanotube array is immersed in a mixed solution containing to-be-detected protein and ionic liquid [P6, 6, 6, 14] [FuA], and the SERS active substrate is prepared. The ionic liquid [P6, 6, 6, 14] [FuA] is introduced into the SERS active substrate, the interaction between the protein and the substrate is enhanced, the ionic conductivity is improved, the overall polarizability of a detection system is increased, and the SERS performance of the protein is improved.
