The invention discloses a method for extracting and purifying zinc-protoporphyrin IX in a nitrate-free dry-cured ham. The method comprises the following steps: removing fat from a biceps femoris muscle sample of the dry-cured ham, and performing vacuum freeze drying; the method comprises the following steps: taking a biceps femoris muscle sample after freeze drying, and carrying out crude extraction on zinc-protoporphyrin IX in the sample by using a 90% acetone aqueous solution; diluting the crude extract with distilled water according to a ratio of 1: 1 (v/v), filtering with filter paper, and adding a sample to a reverse C18 solid-phase extraction small column for purification; and collecting the purified eluent, removing acetone in the purified eluent by using a nitrogen blowing concentrator, and transferring the purified eluent into a vacuum freeze dryer for freeze drying to obtain a zinc-protoporphyrin IX purified sample.
