Zusammenfassung: | The invention discloses a proximity protein marking system ScPhastID with high credibility and high speed and a marking method. A biotin labeled enzyme PhBPL * with high catalytic activity is split into two parts without activity by utilizing a protein splitting technology, namely PhN32 protein and PhC33 protein, the amino acid sequence of the PhN32 protein is as shown in SEQ ID No.2, and the amino acid sequence of the PhC33 protein is as shown in SEQ ID No.3. The biotin labeled enzyme PhBPL * has high catalytic activity. Wherein the smaller fragment PhN32 is fused with the researched target protein as a protein tag, the PhC33 and the PhN32 can be self-assembled to recover the labeling activity, and the PhN32 and the PhC33 form an adjacent protein labeling system ScPhast ID. In the ScPhastID, the positioning influence on the target protein is reduced by using the small volume of PhN32, the marking accuracy is improved, and meanwhile, the marking effect is controlled by controlling the expression of PhC33.
