According to the preparation method, 2-(benzyloxy)-1-(2, 4-bis (benzyloxy)-6-hydroxyphenyl) ethyl-1-ketone is taken as an initial raw material and reacts with 1-bromo-3-methylbutyl-2-ene under the action of a catalyst, and the novel anhydroicaritin intermediate 2-(benzyloxy)-1-(2, 4-bis (benzyloxy)-6-((3-methylbutyl-2-ene-1-yl) oxy) phenyl) ethyl-1-ketone is obtained. Meanwhile, the invention provides a method for preparing anhydroicaritin by using the novel intermediate compound, which comprises the following steps: carrying out rearrangement reaction on 2-(benzyloxy)-1-(2, 4-bis (benzyloxy)-6-((3-methylbutyl-2-ene-1-yl) oxy) phenyl) ethyl-1-ketone, reacting with 4-methoxybenzoyl chloride, cyclizing the obtained product, and debenzylating to obtain anhydroicariin.
