The invention relates to the technical field of abelmoschus manihot flower separation and analysis, and discloses an abelmoschus manihot flower total flavone determination method, which comprises: (1) using hyperoside as a standard substance, using an ethanol solution to achieve a constant volume, and preparing a standard substance solution; (2) measuring the maximum absorption wavelength by using the standard substance solution prepared in the step (1) and a total flavone extracting solution in the abelmoschus manihot flower as a test solution; (3) preparing a hyperoside standard solution with gradient concentration, measuring the absorbance at the maximum absorption wavelength, and establishing a standard curve; and (4) taking a total flavone extracting solution in the abelmoschus manihot flowers, diluting, measuring the absorbance at the maximum absorption wavelength, substituting the absorbance into the standard curve obtained in the step (3), and calculating the concentration.
