The invention provides a primer and probe design method for miRNA detection, a detection composition comprising a primer and a probe designed by the primer and probe design method, and a detection kit comprising the detection composition. By using the primer and the probe designed by the method disclosed by the invention, miRNA detection can be carried out by adopting the one-step method, non-specific amplification is avoided, the sensitivity is high and reaches 1000 copies/mL, and the specificity is good.
