Zusammenfassung:The invention discloses a suspension domestication method for high-titer lentivirus production based on HEK293T clone screening, which comprises the following steps of: S1, carrying out clone screening of HEK293T adherent cells suitable for lentivirus packaging: screening out a plurality of first clone cells with higher infection efficiency, carrying out enlarged culture and building a library; S2, carrying out rapid suspension domestication on the screened HEK293T adherent cells to obtain HEK293T cells which are 93-98% in cell viability, good in cell dispersity and free of obvious aggregation and are successfully subjected to suspension domestication; and S3, screening suspension HEK293T high-titer lentivirus packaging clone cells: screening a virus supernatant with high infection efficiency, and obtaining corresponding clone cells, namely the high-yield suspension HEK293T cell clone of the lentivirus.
