The preparation method comprises the following steps of: cutting Wharton's jelly into tissue blocks with the size of not more than 2mm < 3 >; digesting the tissue blocks at 37 DEG C to 39 DEG C for 30min to 60min by adopting collagenase with the concentration of more than or equal to 1 percent; carrying out mesenchymal stem cell culture on obtained suspension liquid and digested tissue blocks respectively to obtain the umbilical cord mesenchymal stem cells.
