Zusammenfassung: | The invention provides a synthetic method of a dominant malachite green hapten and application of the hapten, and the synthetic method comprises the following steps: coupling 4-formyl phenylboronic acid and acrylic acid under rhodium catalysis to obtain 3-(4-formyl-phenyl)-propionic acid, reacting the compound with N,N-dimethylaniline in the presence of zinc chloride to obtain a recessive malachite green hapten, conducting oxidizing with tetrachloro p-benzoquinone to obtain a dominant malachite green hapten, and cooupling the hapten with carrier protein to obtain the artificial antigen of the dominant malachite green. Experimental results show that the antibody generated by immunizing animals with the antigen is high in specificity and sensitivity, the titer of the obtained antiserum can reach 106, and the detection limit is 0.1 ug/L.
