The invention discloses a sheep hypertonic complexing agent semen dilution preservation solution which is simple and convenient to prepare and an application method thereof. The preservation solution is prepared from 2.4 g of trisodium citrate dehydrate, 1.0 g of glucose, 0.8 g of urea, 0.05 g of KCl, 0.02 g of EDTA-Na2 and double distilled water with the constant volume of 100 mL.
