The method comprises the following steps that in-vitro amplification is carried out on annular plasmids with tos in a PCR amplification mode, enzyme digestion is carried out by using TelN protelomerase to obtain a linearized cloning vector, and meanwhile, sequences which are the same as side wings of a target genome segment are introduced to two ends of the vector to obtain a high-concentration linearized vector; in-vitro assembly is carried out on the linearized vector and the genome library to obtain an assembled product; a TelN protein expression gene is integrated into a host gene to obtain a host integrated with the TelN protein; and the assembled product is transferred into a host integrated with the TelN protein, and the target fragment is screened.
