Zusammenfassung:The invention belongs to the technical field of primary cell separation and culture, particularly relates to a low-cost in-vitro separation and culture method for primary mouse spleen reticulated fibroblasts, and solves the problems of very high separation cost, very complex separation technology, high separation technology threshold, long culture period, poor stability and the like of FRCs cells in the prior art. The low-cost primary mouse spleen reticular fibroblast in-vitro separation and culture method comprises the following steps: digesting mouse spleen fragments in one step by adopting collagenase P, and digesting and separating to obtain mixed cells; carrying out in-vitro culture on the mixed cells, and digesting by using trypsin containing EDTA; and gradually purifying the mouse spleen reticular fibroblasts for multiple times until the final purity is analyzed and identified to be more than 98%. The method is simple and convenient to operate, low in cost and low in equipment requirement, and does not
