The invention discloses an enzymatic preparation method for preparing glucosamine from glucose and belongs to the technical field of bioengineering. According to the method, the glucosamine is prepared by taking the glucose as a substrate; the method comprises the following steps of: combining biological enzymes, or an expression vector or a cloning vector for expressing the combined biological enzymes, or a transgenic cell line for expressing the combined biological enzymes, or a gene engineering bacterium for expressing the combined biological enzymes; and preferably, combining four types of biological enzymes including hexokinase, 6-phosphoglucose isomerase, glucosamine synthase and glucosamine 6-phosphodephosphatase.
