The system comprises a liquid phase combination pump, a mixer, a sample introduction device, a separation column and a detector which are connected in sequence, the liquid phase combination pump can independently pump at least three solvents to the mixer as mobile phases, the three solvents are respectively a weak solvent, a strong solvent A and a strong solvent B, and the elution capacities of the weak solvent, the strong solvent A and the strong solvent B on a separated target object are sequentially enhanced; the liquid phase combination pump forms a first elution gradient and a second elution gradient of a mobile phase by adjusting the ratio of pumped solvents, the first elution gradient is the ratio of the weak solvent to the sum of the strong solvent A and the strong solvent B, and the second elution gradient is the ratio of the strong solvent A to the strong solvent B.
