The meningococcus culture medium comprises a solution A and a solution B, and a volume ratio of the solution A to the solution B is (8.5-9.5):(0.5-1.5). The preparation method comprises the following steps of: (1) weighing acid hydrolyzed casein, yeast extract powder, L-cystine, KH2PO4, Na2HPO4, glycine, L-sodium glutamate, NH4Cl and NaCl according to the preparation amount, putting the raw materials into a container, adding water for injection to dissolve the raw materials to obtain a solution A; (2) weighing MgSO4, MnSO4. H2O and glucose according to the preparation amount, and adding water for injection to dissolve, so as to obtain a solution B; and (3) respectively sterilizing the solution A and the solution B, and mixing them according to the ratio. The culture method of meningococcus comprises the following steps of
