The invention discloses a high-sensitivity method for determining the cAMP concentration in a biological fluid sample by liquid chromatography-mass spectrometry. The method comprises the following steps of: (1) pretreating a to-be-determined sample; (2) preparing a standard curve through liquid chromatography-mass spectrometry detection; and (3) detecting the concentration of cGMP in the to-be-determined sample by liquid chromatography-mass-spectrometry.
