The method comprises the following steps of: 1, preparing a to-be-detected object into a glass slide, and carrying out qualitative identification on phytoplankton algae in a visual field; and 2, drying the glass slide, and after drying, carrying out the counting process of the phytoplankton algae and the whole-piece counting process of the rare algae under a glass slide microscope.
