Zusammenfassung: | The invention discloses a novel creation method of self-incompatible brassica napus germplasm, and relates to the technical field of plant genetic engineering. The method mainly comprises the following steps of (1) identifying homologous sequences of two SMI2 in a recessive affinity brassica napus genome, wherein the homologous sequences are named as BnS6-SMI2 and BnS7-SMI2 respectively, and the nucleotide sequences of the homologous sequences are shown as SEQ ID NO.1 and SEQ ID NO.2 respectively; and (2) designing three sgRNAs based on CRISPR/Cas9 according to the specific sequence of the BnS6-SMI2 gene, wherein the nucleotide sequences of the sgRNA1, the sgRNA2 and the sgRNA3 are shown as SEQ ID NO.3, SEQ ID NO.4 and SEQ ID NO.5. The method proves that the BnS6-SMI2 regulates the self-incompatibility, and creates strains of different mutation types by using CRISPR/Cas9, and the self-incompatibility of the BnS6SMI2 is obviously lower than that of a receptor material 326; and the selfing progeny of the brassi |
