Zusammenfassung:The invention relates to a construction method of yeast site-directed double-DSB synchronous induction model. A GAL-I-SceI-KanMX transformation cassette is integrated into the genome of Saccharomycescerevisiae by a chemical transformation method to form the model. According to the invention, I-SceI endonuclease is expressed by activating a promoter and recognizes I-SceI recognition sites at bothends for cutting, so that the generation of two DSBs with different intervals can be synchronously induced, and interference of other damage types is eliminated. More importantly, the model can be used to study the synchronous generation of "aggregation" DSBs, which is a characteristic damage type generated by cancer radiotherapy and numerous chemotherapeutic drugs, and this model is a powerful tool for studying cell signal pathways and drug toxicology.
