The preparation method comprises the following steps of (1) performing enzymolysis: heating slurry obtained by grinding soybeans, then cooling the slurry, adding composite proteases, performing heat-insulating enzymolysis, then performing filtering, and performing centrifugation to obtain a soybean peptide solution; (2) performing fermentation: heating the soybean peptide solution, then cooling the soybean peptide solution, performing inoculating with zymocyte, performing temperature-controlled fermentation, performing centrifugation to obtain supernatant and precipitate, performing vacuum concentration and spray drying on the supernatant to obtain peptide powder, performing freeze drying on theprecipitate to obtain active bacterium powder, and uniformly mixing the peptide powder with the active bacterium powder, to obtain the soybean peptide rich in active lactobacillus.
