The invention discloses a polyionic liquid modified nano material, a preparation method thereof and application of the material in enrichment of phosphorylated peptide in a sample containing phosphorylated peptide. The preparation method comprises the following steps: (1) polymerizing a vinyl-modified substrate material, vinyl and organic phosphonate functionalized ionic liquid to obtain a polyorganic phosphonate ionic liquid functionalized material; and (2) acidifying and hydrolyzing the polyorganic phosphonate ionic liquid functionalized material, and then combining with metal ions to obtainthe affinity material of the polyorganic phosphonate functionalized ionic liquid.
