The method includes:establishing a multi-stage ship lock chamber water supplementing calculation model; the operation stage number of the ship lock, the upstream and downstream positions and the initial water levels of the lock chambers serve as input of the model, the high operation water level and the low operation water level needed by all the lock chambers for meeting navigation operation conditions are calculated, whether the lock chambers need water supplementing or not and the needed water supplementing thickness in the navigation process are judged according to the model, and automatic operation of the ship lock is facilitated.
