The invention discloses a method for detecting a particle size of recombinant enterovirus 71 type virus-like particles and an application thereof in preparation of hand-foot-and-mouth disease vaccines. The method comprises the following steps: adding polysorbate 80 into a to-be-detected recombinant enterovirus vaccine 71-type virus-like particle sample, and uniformly mixing until the final concentration of polysorbate 80 in the sample is 50 [mu]g/mL-400 [mu]g/mL; and detecting the particle size of the virus-like particles in the sample added with the polysorbate 80.
