The invention discloses a tissue culture rapid propagation method inducing seedling formation from hairy roots of Saccharum officinarum L. The tissue culture rapid propagation method sequentially includes the following steps that (1) the stem tips of the Saccharum officinarum L. are acquired and grafted, specifically, the mature stem tip sections of the Saccharum officinarum L. are cut off at 9:00-10:00a.m. of a continuous bright sunny day, external-layer tissues are stripped, and immature stem tips are taken to carry out grafting; (2) the hairy roots are induced by explants; (3) the hairy roots induce callus; and (4) the callus induces seedlings. Culture mediums with different compositions and ratios are used in the steps. By adopting the tissue culture rapid propagation method, sterile seedlings of the Saccharum officinarum L. can be quickly obtained, and the tissue culture rapid propagation method is beneficial to industrial production; and the induction rate of the hairy roots of Saccharum officinarum L.
