Zusammenfassung: | The invention discloses an effective method for extraction and purification of an endoplasmic reticulum and protein thereof from plant leaves. The method adopts plant leaves as the materials, by meansof adjusting the material type, the material dosage, the grinding mode, the centrifugal rotor type, the protein resuspension mode and the other processes, and through combination with differential centrifugation method, high-quality plant leaf endoplasmic reticulum protein can be obtained. Plant leaves at an appropriate age of about 15 days are adopted, a mode of directly adding a buffer solutioninto the fresh leaves for grinding is adopted to replace a mode of ultrasonic extraction after liquid nitrogen grinding, a constant-angle rotor is replaced by a horizontal rotor during centrifugationat a rotating speed of 12000xg, and compared with total protein, the positive markers GRP78 and CNX of the purified endoplasmic reticulum protein are respectively increased by about 3.3 times and 6.1times, and chloroplast posi |
