The invention provides a screening method of activator of oil reservoir petroleum hydrocarbon degrading bacteria and a method for evaluating activation effect. The screening method comprises the following steps of screening the types of nitrogen sources and/or phosphorus sources by adopting a single-factor method; preparing a to-be-selected activator by using oil reservoir formation water, whereinthe activator comprises a carbon source, a nitrogen source, a phosphorus source, magnesium sulfate, calcium chloride and yeast powder; performing oxygen-limited culture, and shaking and culturing ata target oil reservoir temperature for 2-4 d at 100-200 rpm to obtain activated fermentation liquor; and measuring OD600, emulsifying capacity and/or surface tension of the activated fermentation liquor, and screening the activator with effects of directionally regulating and controlling degradation function bacteria of the petroleum hydrocarbon in the oil reservoir according to a measurement result.
